The process took 1 day. I interviewed at Lowe's Home Improvement in Apr 2009
Interview
Was contacted 3 days after applying and told to come in for a interview 2 days later. Went there and met with the HR manager had initial interview with her then met with department manager then store manager. Some questions asked caught me off guard like what would you describe your greatest weakness to be, other then that it was typically easy and the managers I met were nice they gave me a drug test which i had to hold a stick in my mouth then the interview was over they called me few days later and told me when to come in and start. The pay was better then i had before but not negotiable.
I applied online. The process took 3 days. I interviewed at Lowe's Home Improvement (Cary, NC) in Feb 2009
Interview
HR manager interview, then interviewed with the store manager. Pretty basic questionaire that both are reading from and scoring you right in front of you. Not too stressful because you can tell they aren't really interested in what you're saying. Totally depends on who you end up interviewing with though, I've worked in a few different stores and the level of professionalism among managers varies greatly.

I applied online. The process took 3 days. I interviewed at Lowe's Home Improvement (Plover, WI) in Mar 2010
Interview
I applied probably two weeks before I got a call to set up an interview. I went in to the first interview with a wonderfully nice woman (HR) who asked the typical 'tell me about yourself' questions. After that, we set up an interview for two days later. I went through two more people that day and the final interview took only 5 minutes. He said 'we're going to make you an offer', and I gladly accepted.
It was all one on one interviews, very short too..So don't get nervous
